It may not be completely explained in words how fortunate I am to be a graduate student of Osaka University, Department of Material and Life Science, Laboratory of Molecular Biotechnology under supervision of Prof. Dr. Shigenori Kanaya and other heart-warming staffs. I am Nujarin Jongruja from Thailand. I am currently studying in 1st year of Doctor Course. I gratefully received an opportunity to go abroad for internship at Laboratory of Molecular Genetic, National Institute of Health, Maryland, USA for about 1 month from Jan, 12 to Feb, 14 2010. Dr. Robert J Crouch, the head, has been interested in processes involved in cellular DNA replication and the relationship of HIV replication to cellular DNA replication events and how to use the resultant information for therapeutic purposes. My assigned work during the internship is to construct a yeast-RNase H1 containing plasmid with the antibiotic selectable marker. This plasmid can be expressed both in E. coli and yeast with the selectable markers of ampicilline-resistant and URA3 genes marker, respectively. However, we would like to replace URA3 gene with the kanamycin-resistant gene, which causes the resistance of G418 in yeast system instead of kanamycin-resistance. Recombinant-mediated PCR-directed plasmid construction technique was applied in order to insert kanamycin-resistant gene. The techniques that I have learned during the internship are different from what I get used to because, in our laboratory, most techniques are for DNA manipulation and protein overproduction in E. coli. Therefore, this is a great opportunity to know the techniques for eukaryotic cells. Beside the academic experiences, I agree with the idea that the more you see the world, the more you see your own defects and improve yourself. Several good attitudes and behaviors of American and Japanese people differ from mine. I hope and try to adapt those to be a better me.
